Test Panels

Alanine Aminotransferase (ALT)

Overview of Test

Alanine aminotransferase is an enzyme that is located exclusively in the cytosol of liver cells and participates in modifying amino acids. Often tested in conjunction with aspartate aminotransferase. ALT is one of the several tissue enzymes that catalyze the interconversion of amino acids and oxoacids by transfer of amino groups. Aminotransferase activity reflects the rate at which enzymes enter and are cleared from the circulation. Assists in the diagnosis of acute liver damage.

Grossly hemolyzed. Frozen in gel tube. Specimen quantity not sufficient. Not centrifuged.

Methodology

ALT catalyses the transamination of L-alanine and α-ketoglutarate to produce pyruvate and glutamate. Pyruvate produced is then measured with LDH and the conversion of NADH to NAD. The rate of disappearance of NADH and the resulting decrease in absorbance, measured bichromatically at 340 nm, is proportional to the ALT activity.

Collection Instructions If Written

After draw, allow the specimen to clot by letting it sit at room temperature for approximately 30 minutes. Specimens should be centrifuged within 2 hours of collection. Centrifuge the specimen at 3,400 RPMs for 15 minutes to separate the red blood cells from the red blood cells from the serum.
