Test Panels

Alkaline Phosphatase

Overview of Test

Quantitative analysis of alkaline phosphatase levels. This is of interest in the diagnosis of liver disorders and bone disease associated with increased osteoblastic activity. Moderate increases of ALP may be seen in several other conditions such as Hodgkin’s disease, congestive heart failure and bacterial infections because it is nonspecific and from many tissues.

Causes For Rejection

Grossly hemolyzed. Frozen in gel tube. Specimen quantity not sufficient. Specimen not centrifuged.

Methodology

Beckman Coulter AU680 analyzer with Carolina Liquid Chemistries reagent. Alkaline phosphatase hydrolyses p-nitrophenyl phosphate to form p-nitrophenol, the rate of increase of which is proportional to alkaline phosphatase activity measured bichromatically at 410/480nm.

Collection Instructions If Written

After draw, allow the specimen to clot by letting it sit at room temperature for approximately 30 minutes. Specimens should be centrifuged within 2 hours of collection. Centrifuge the specimen at 3,400 RPMs for 15 minutes to separate the red blood cells from the serum.
